Development of High Efficient New Production Process for Proteins

Sanyo Chemical has developed the high efficient production process for proteins by applying our strong points of surfactant technologies. We realized extracellular production of recombinant proteins by Escherichia coli ( E.coli) with high productivity, which has not been impossible hitherto. E.coli is a bacterium used as host for intracellular expression of heterologous proteins in biotechnology generally. Our process enables to improve the productivity of proteins and extends the possibilities of industrial use of the valuable proteins which have not been obtained enough in industrial scale so far.

 

Conventional problems
Generally it is well known as the production method for proteins by expression of the target in E.coli and taking it out after destruction the cell membrane. E.coli is applicable and versatile for many kinds of proteins. But its production capacity is limited because the target protein is expressed only inside the cell and never secreted outside. Also its purification methods are complicated because there are many contaminants through destruction of the E.coli cell, to take out the protein.
The other method is also available that is producing extracellular proteins by using Bacillus subtilis or Aspergillus oryzae. This process is efficient because the target protein is continuously secreted outside. However its versatility is extremely low because the proteins which can be applied to are limited. In addition, the E.coli, the most versatile bacteria for production of recombinant proteins, is conventionally inadaptable for this method.

 

Abstract of the new technology
Our development realized extracellular production of recombinant proteins using E.coli by applying our strong points of surfactant technologies. We have found the special surfactant, which has an effect for E.coli’s cell membrane to be more permeable. This enables to establish the new extracellular producing process of the proteins using E.coli. Our process extremely improves the productivity for proteins by being continuously secreted to outside the cell and expressed inside. And purification becomes efficient by less contaminants derived from destruction of the cell. Because of that, our development extends the possibility of the industrial use of the valuable proteins which have not been obtained enough in industrial scale so far.

 

Fields of application
Our development improves the productivity of proteins existing in the market. Furthermore, it extends the possibility of the industrial use and broad fields applied to, for the proteins having been produced extremely in small amount so far. We expect that our technology contribute to the many fields such as pharmaceuticals, detergents, cosmetics and process foods, etc. thorough developing the effective proteins.

 

Future plans
We have constructed about a 100L test facility on laboratory scale and started the several collaborative studies for the industrial production.
We are trying to assist the productions of various proteins through flexible corresponding to various needs by collaborative studies, entrusted productions, license contracts and others, with not limited to chemical and pharmaceuticals industries but including universities and research organizations.
